تلعب إستيلاء على المركبات الكبرى 5 من منظور الفرد الأول و الثالث على بلايستيشن 4 و إكس بوكس ون ومايكروسوفت ويندوز، تلعب بمنظور الفرد الثالث علي بلايستيشن 3 واكس بوكس 360 وتجمع مكونات قيادة المركبات والأكشن في عالم مفتوح، تسمح للاعبين بالتفاعل مع العالم كيفما شاءوا. تقع فعاليات اللعبة في ولاية سان أندرياس الخيالية، المبنية على في جنوب كالفيورنيا، تضمن لللاعب التجوال بحرية في مناطق الريف ومدينة لوس سانتوس، المبنية على لوس أنجلوس. طور الحكاية يروي بواسطة ثلاثة شخصيات يتم التحكم بها ويقوم اللاعب بالتحويل فيما بينهم، وهم: مايكل وتريفور وفرانكلين. الحكاية تتبع أتعاب الثلاثة الذي يقومون بالسطو على المصارف بهدف الحصيلة. يبقى في اللعبة طور اللعب الجماعي، والذي يسمح إلى ستة عشر لاعب بالمساهمة في اللعب التعاوني والتنافسي.

إستيلاء على المركبات الكبرى 5 "تطور جميع الأشياء" متواجد في الأجزاء الفائتة في السلسلة. عالم اللعبة أضخم من عالم ريد ديد ريدمبشن وغراند ثفت أوتو: سان أندرياس وغراند ثفت أوتو 4 مجتمعين. الأنحاء في اللعبة تشمل الشواطيء والجبال والبراري ومنطقة تشبه "سالتون البحر" في كاليفورنيا، وقاعدة عسكرية والضواحي. المحيط مصمم بالكامل وقابل للأستكشاف.[7]

التليفون المحمول الذي ظهر في إستيلاء على المركبات الكبرى 4 سيرجع إلا أن العديد من خصائصه قد تغيرت. يستعمل التليفون المحمول في الأساسً في النشاطات. ومن الممكن أستخدامه ايضاً للدخول إلى إنترنت اللعبة. سوف يكون هناك نسق أقتصادي "خافق بالحياة ومرح"، إلا أن اللاعب لن يمكنه شراء المنشآت كما في فايس سيتي و فايس سيتي ستوريز. نسق التصويب تطور "بشكل ملحوظ" من حيث الإحساس والميكانيكية، ونظام القتال قد تطور ايضاًً.[9]
